Rp: Removal of the mismatches base pairs

DNA polymerases increase the accuracy of Replication by selecting the correct base for insertion and by proofreading the newly synthesized DNA to remove mismatched bases. This proofreading activity is responsible for the high fidelity of DNA polymerase.
The erroneous base is removed by the activation of the 3' -> 5' exonuclease activity of DNA polymerase. If a 5'-deoxynucleotide is not complementary to the corresponding base of the template strand, the enzyme reverses its synthesis activity and hydrolyzes the phosphodiester bond formed between the primer and the erroneous base.
