SLE is supposed to have antibodies against the smrnps or histones.....now the big q...how does an antibody specific for smrnps or histones bind to a cell surface at all seeing that it should all be in the nucleus & not on the cell membranes????

Systemic lupus erythematosus (SLE) is a chronic autoimmune disease characterized by the production of autoantibodies to a spectrum of nuclear antigens. (Key word spectrum) Sle1 mediates the loss of tolerance to nuclear antigens; Sle2 lowers the activation threshold of B cells leading to polyclonal IgM antibody production and increased numbers of B1 cells; and Sle3 mediates a dysregulation of CD4+ T cells that potentiates polyclonal IgG antibody production and decreases in activation-induced cell death in CD4+ T cells. ...results indicate that Sle1 is key for the development of fatal lupus in this model, indicating that the loss of tolerance to chromatin mediated by Sle1 is essential for disease pathogenesis and identifying Sle1 as a strategic target for therapeutic intervention in SLE. also you might want to ck out: casciola-rosen la, anhalt g., rosen A. I think what you asking is how the AB is formed id those things are inside cell??? If so... At any given time pices of DNA and all cell nuclear components are in circulation from cells that undergo appotosis or cell mediated immunity or other lytic cell "stages". So immune system sees those particles and makes AB to them. A lot of us have those AB, but not have the Dx, nobody knows why. But that makes those AB non-specific, unless most other 4 out of 12 signs are present. Hope this helps.
