How does it cause fatty change in the liver? As in mechanism of fatty change??

Its ok, basically CCl4 is converted to CCl3 radical by P450 which is a highly toxic molecule. It diffues through the cell membrane and causes lipid peroxidation of the membrane. Ribosomes disaggregate resulting in decreased protein synthesis, meaning to translation, leads to no formation of apolipoproteins causing accumulation lipids and fatty change. It also damages plasma membrane from products of lipid peroxidation which leads to cellular swelling, calcium influx into the cell, mitochondrial damage, and eventually cell death.

Angiosarcome also called hemangiosarcoma, is a malignant tumor of the liver associated with toxic exposure to polyvinyl chloride, thorotrast, and arsenic.
Can anyone talk about effects of acetaminophen on liver??

acetaminophen causes necrosis of liver & renal tubules.
mechanism is free radical damage.

yeah , toxic metabolites of PCM bind to SH gps on n acetylcystein thus releasing glutathione.
should be given within 10 hrs of overdose.

n acetylcystein conteins sulfhydryl groups, which are bind to toxic metabolite ( n acetyl benzo-something). Must be administered within 10 hrs of the overdose.

I read recently that SAM has been proved to work better than CoX inhibitors in arthritis.
