CD 40 is involved in isotype switching, and binds CD40 on B cells and is a marker on T cell. CD 28 is also a marker on T cell, and is a costimulatory molecule that is required for activation of T cells. It binds b7 on B cells.

CD 28 on T cell...interacts with CD 80/86 on B-cell. results in T cell actication and differentiation. CD 40 on B-cell, interacts with CD40 ligand (also known as CD 154) on T cell , isotyoe switch enabling.
