how does it contribute to cancer development ?

Tumour suppressor genes halts progression of cells for G1 phase to S phase, i dont know which tumour supressor gene you are talking about. They may induce apoptosis.

both 1 ans 2 can lead to cancer

:arrow: protooncogene---> amplification /induction/ gain of function ---> cancer .
