The initial vaccine against H. influenza prepared with a capsular polysacch was not effective. Subsequent vaccines have improved by conjugation to proteins. Why does conjugation improve immunogenicity?

a) CD4 is activated
b) CD8 response is increased
c)direct activation of Bcells by vaccine
d)nonimmune clearance of vaccine decreased
e) phagocytosis of vaccine increased


Does anyone know a site that can explain immune response?

I am confused because only live viral vaccines can cause a cell mediated response so how do conjugated proteins cause an immune response? Don't they activate Bcells?!!!

or does cell mediated response just mean that the (infected) cell causes an immune response?

The limitation of polysaccharide vaccines is that they are enable to activate Th cells and trough them to active response of Bcell and produce Ig ( what we want in vaccination ). That is why the polysaccharide vaccine is conjugated with some sort of protein carrier, like in Hib vaccine, which result in higher imunogenicity than polysacch. alone.

A.. T cell dependent Antibody repsonse is increase with conjugation.

Carbohydrates activate b cels
but proteins bind to HLA 2 which bind CD 4 of T-helper to make memory by activating TH2 and causing class switch via CD 40L ( lymphoctes ) - CD40 ( B cells )
