When an inflammatory process has to start somewhere , leukocytes escape out of blood vessels to reach that site. You know , the leukocytes normally flow in the center of the vessel. To get out of the vessel wall , first of all they move closer to the wall, this is called margination.Leukocytes tumble on the endothelial surface , transiently sticking along the way. This process is called rolling. 2nd step is adhesion of the leukocytes to the endothelium. This is where selectins and integrins play their role.
Selectins are expressed on leukocytes and endothelial cells. P and E selectins on the endothlial molecules and L selectin on the surface of most leukocytes. There are other receptors called Sialyl-Lewis-X on the leukocytes.
Now while the leukocytes are rolling along the endothelium , their Sialyl-Lewis-X receptors attach to the E and P selectins on the endothelium and their L selectins attach to L selectin receptor on the endothelial molecules and leukocytes are loosely attached to endothelium.
Now there are some other glycoproteins on the leukocytes called integrins and their receptors on the endothelial moecules called ICAM1 and VCAM1 . Integrins are normally present in the non activated form. Once the selectins are bound to their receptors , these integrins are activated and bind to VCAM and ICAM on the endothelium . This way leukocytes are firmly bound to endothelium so that diapedesis can take place.
So , one liner answer to your question is , selectins and integrins play role in adhesion. Selectins for loose adhesion and integrins for tight adhesion.
