methotrexate is competitivly inhibit the enzyme dihydrofolate reductase DHFR which have an important role in folic acid synthesis which needed for synthesis of purine and pyrmidine especially thymidine so it affects the s phase

yes METHOTREXATE IS CYTOSTATIC DRUG a S-phase selective and to add more to "inb6des's post -----"methotrexate is competitivly inhibit the enzyme dihydrofolate reductase DHFR which have an important role in folic acid synthesis which needed for synthesis of purine and pyrmidine especially thymidine-------------->REQUIRED FOR DNA SYNTHESIS therefore, inhibits the synthesis of DNA, RNA, thymidylates, and proteins!
