Atypical, or reactive lymphs, don't always show up with Mononucleosis.

We use a rapid test for Mono that can be done in 5 minutes with one drop of serum. I believe it's an antibody test but I've never read the package insert :-(. Rapid mono tests today are much nicer than the ones I trained with 30 years ago.

Bactitech, you are correct . Monospot is testing for presence of Heterophile AB, that reacts with sheep's RBCs. Fast and cheap. Atypical Lymphs though present, are not specific nor are they present all the time. If virus is not shedding, cells are not showing up.
