-GI side effects due mainly to inhibition of cox-1 -anti-inflam therapeutic effect due mainly to inhibition of cox-2 THUS COX-2 is out in the market. some cox-2 inhibitors proven to increase risks of thrombosis and embolism High doses of some NSAIDs also increased risks of thrombosis and embolism mediated by inhibition of cox-2 Antiplatelet effect of aspirin due largely to inhibition of cox-1 and there is less effect on inhibition of cox-2, so aspirin does not lead to increase risk of thrombosis??? In selecting the drugs, need to balance the risk of cox-1 inhibitors- in causing GI problems vs the risk of cox-2 inhibitors- in causing thrombosis Pls correct me if I am wrong
