I chose C since you actually see the genetic codes by nucleotide sequencing. I'm pretty sure it isn't D since that usually is just amplifying the DNA ... not reading the DNA. As for A, I'm not sure how to rule it out other than arguing that you are dependant upon creating lots of probes to determine if the 2 are the same vs just sequencing (so this is less efficient). Also, I think usually labs will use hybridization/southerns to check but the confirmatory stage is via sequencing.

Ill go answer-c since a nucleotide is {a base , sugar and a phosphate} if you can sequence the nucleotide I think that would do it.

yeah I go with C , I did not see it. Hope never make this mistake in the real exam.
