Vd = Dose/C0

C is the amount of drug in the plasma at zero time.

What i don't get is that Shouldn't dose equal to C, since dose is the amount of drug u infuse into the patient??? Some one plz help me out here. thanks

Vd= Amount of drug in the body / plasma drug concentrration........................... do you thing that all the drug that we give is present in the plasma at zero time? Can be in IV form but not for others route

D is the amount of drug(dosage) given -could be PO in milligrams or IV -30cc per hour

C is the plasma conc. at time 0- its the non- ionized part that has the effect.

[most drugs will exhibit 1st order kinetics]

Part of the dose administered gets out of the blood/plasma, for exmple to target tissues. I think that's the main reason for difference between dose and plasma concentration.

also there is absorption, distribution, metabolism and even excretion of the drug. not all is in the plasma. some can be sequestered also
