Deletions are routinely detected by two methods: Southern blotting (using a probe from either a cDNA or genomic library) or polymerase chain reaction (PCR) using primers specific for the gene. Both of these techniques will show size changes that will allow the investigator to determine the size of the deletion. Deletions may sometimes be detected by using restriction enzymes; for example, the loss of a normally present restriction site may indicate deletion of the DNA in the vicinity of that site.
