Actively dividing fibroblasts in culture are transfered from a 37C incubator to a 0C ice bath, and 10-6(power) M colchicine is added to the culture medium. After 30 min, many cells have een arrested in mitosis. Which cytoskeletal protein is most likely to be involved? A. Actin B. Desmin C. Dynein D. Keratin E. Kinesin F. Lamins G. Myosin H. Neurofilament proteins I. Tubulin

I? I have just remembered that Colchicine, Vinbristine, Vinblastine act on tubulin.

I remember Colchine acts on microtubule and we know that microtubule consists of Dyenin and Kinensin So ing can u please put the right answer

tubulin that forms the spindle fibers cannot get polymerized due to colchicine... leading to arrest in metaphase of the M phase of cell cycle

surgery, this is an NBME q and does not have an answer!! waqastariq, i thought mitosis was arrested in anaphase by colchicine

As far as I know mitosis is arrested in metphase

well surgery, i am probably the last person to discuss this topic but here is what i can understand. the major component of microtubules are tubulins (alpha and beta) they have 2 motors, dyein and kinesin. dyein causes movement towards negative end while kinesin causes movement towards positive end. in metaphase and anaphase, both kinesin and dyenin are involved. colchicine to my knowledge acts on the tubulin itself and not the motor. colchicine and vinca alkaloids bind to the spindle and prevent polymerization of the tubulin which inibits the spindle and arrests the cell in mitosis.

and for the sake of completion here r 2 relevant posts i liked on this toopic PacKliTAXEL--> TAXANES keep the spindle PACKED, prevents spindle disassembly. Vincristine and vinblastin Believe in ONE( V for one) tubulin i.e prevent assembly spindle assembly by inhibiting polymaerization of tubulin Dimers (though) these are obviously CCS (cell cycle specific) for the M phase ..
