I think of it this way... Splicing site is the boundary between exons and introns -> which are going to be cleaved by snRNA or spliceosome at this specific boundary which is recognized by its highly specific sequence.. This is the last process of hnRNA for becoming mRNA. In this particular example theyre not showing the 3' end of the whole mRNA just the 3' end of an intron, this specific sequence recognized by snRNA has been lost so the splicing is going to occurr somewhere else, downstream, upstream it depends this would be just a random event..

The mutation would affect splicing. This is because the last codon of the normal intron is a stop codon. If it is mutated to be read as an amino acid codon then the intron will not be spliced at the 3' end and will be incorporated into the mRNA.
