If the inserts are ligated into a Pstl restriction site in the erythromycin resistance gene of the plasmid, the entire populatin of transformed bacteria should be first be grown in media containing

A, both chloramphenical and ampicillin

B, both chloramphenical and erythromycin

let me explain. if it is treated with chloramphenicol the cells without the resistence plasmid vector will be killed and the cell with plasmid will stay.

if treated with erythromycin then the resistance to this drug is already disturbed by the insertion so the cells with the vector also die off.

i think u re right, after this do we treat them with erythromycin, bacteria of interest would die. we can blot them, am i right????
