The complete DNA sequence of autosome 21 is being determined by Pstl restriction endonuclease digention of chromesomal DNA, restriction fragment ligatin into a plasmid rector containing both a chloramphenicol resistance gene and erythromycin resistance gene, transformation of E.coli cells with recombinant plasmids, and DNA sequencing of the cloned inserts. If the inserts are ligated into a Pstl restriction site in the erythromycin resistance gene of the plasmid, the entire populatin of transformed bacteria should be first be grown in media containing A, both chloramphenical and ampicillin B, both chloramphenical and erythromycin C, chloramphenical D, erythromycin

let me explain. if it is treated with chloramphenicol the cells without the resistence plasmid vector will be killed and the cell with plasmid will stay. if treated with erythromycin then the resistance to this drug is already disturbed by the insertion so the cells with the vector also die off.
