can anyone pl explain me the mechaninsm of Rb supressor gene n p53 supressor gene in cell cycle??

what i remember is that it works like p53 in that it needs a specific signal before it will allow cell into S phase of cell cycle.

The p53 protein senses DNA damage and can halt progression of the cell cycle in G1. Both copies of the p53 gene must be mutated for this to fail so mutations in p53 are recessive, and p53 qualifies as a tumor suppressor gene.

The Rb gene product interacts with a protein called E2F, nuclear transcription factor involved in cellular replication functions during the S phase of the cell cycle.

Rb if is Hypophosporilated binds E2F and blocks it.

If ther's a mytotic signal, Rb is phosforilated by Cdk4/CyclinD, Cdk6/CyclinD or Cdk2/cyclin E in his N-termino domain and is degradeted. E2f can go into the nucleus and allows trasfcription of Dna pol, tim Kinase. so the cell can progress from G1 to S.

p53 is the guardian of genome. He's activated by stresses like Dna damage and arrestshe cell cycle or leads to apoptosis if damage is too intense.

Normally p53 is degradeted thanks to Mdm2; in presence of stress he goes to the nucles and makes trascription of

-p21

-Gadd45 (Dna repair)

or apoptotic stimuli (bim..)
