The nucleotide sequence encoding four amino acids near the amino terminus of a 10- kd protein is 5´- ACT GAT TGC GTT -3´. This sequence is mutated by insertion of a single nucleotide (A) in the second codon and becomes 5´- ACT GAA TTG CGT -3´. Which of the following consequences of this mutation is most likely?

A ) Altered cell-cycle checkpoint

B ) Decreased rate of transcription

C ) Protein truncation

D ) Single amino acid substitution

C

D. GAT(Asp) for GAA(Glu)

C.(TTG),stop codon.

  #5

does TTG translate into a stop codon???

  #6

Stop codons UAG(TAG), UAA(TAA) and UGA (TGA): you are gone, you are away, you gone away...TTG or UUG is not a stop codon, it codes for Leucine... i think the answer is D. and in some many cases the third position is not revelant (it wobbles)...raised eyebrow

C - a frameshift leads to misreading of all nucleotides downstream resulting in protein truncation

but it is not a wobble since there is an inserted nucleotide

  #8

its a frame shift mutation, leading to truncated protein

  #9

sorry i was wrong

  #10

i also go for c

  #11

oh yup it's C...i got it...

5'ACT GAA TTG CGT 3' -A coding strand

mRNA-5'ACU GAA UUA CGU 3'-new mRNA

original seq 5'ACT GAT TGC GTT 3'

mRNA 5'ACU GAU UGC GUU 3'

As there is no UAA,UAG,UGA -no truncated protein

3 a a substitution

can't change rate of transcription - as no seq of TATA or CAAT inv

ANS- Should be A

  #13

genrally most of the frameshift mutations lead to truncated proteins.

still cant say why it cant be A. but truncation is the rsult of most frameshif mutations

well u took my words msyampgrin.. i was going to type the same, that is that hwnever we have a frameshift mutation, there will always be early truncation of the protein

A new protein as it changes a no. of A.A .-------can lead to stop codon when further sequences are consider.

  #18

yes that is why we have a truncated protein

see you have your answer
