I was noting down key facts in DNA replication & repair for myself & thought I could share this with you. Please let me know of any inaccuracies. add on to this as well ... DNA Replication buzz words 1) DNA replication - if they ask for protein with protective qualities that is damaged or attacked by virus - likely to be ssDNA binding protein ( protects the ssDNA that is formed by helicase)....if they ask for an enzyme that keeps it unwound once converted to ssDNA by helicase - topoisomerase. 2) if bases are mismatched during polymerisation, say naturally, it is 'proof read' or corrected by DNA polymerase which has an added property of exonuclease besides its job of joining bases (nucleotides) back again. remember, RNA polymerase has no proof reading ability, instead make all 3 types of RNA. 3) if the bases (thymine) are destroyed by UV think of 2 words : excision endonuclease and its deficient state - xeroderma pigmentosa. remember this enzyme helps in removing the thymine dimer (pyrimidine base) 4) topo 2 (DNA gyrase) - rapidly dividing cells unwounds the DNA making it loose (negative supercoiling)- key step for cell division drug association - quinolone (norflox) or anticancers 5) normal transcription - DNA to RNA ; reverse transcriptase - RNA to DNA 6) enzymes for DNA repair - think of 2: polymerase & ligase
