I believe it has to do with the substitution of one body part for another. These homeotic genes encode for gene regulatory proteins known as homeodomain proteins. homeotic genes contain a 180 base pair sequence (homeobox) that encodes a 60 amino acid long region (homeodomain) Pax 3 for Waardenburg's syndrome and rhabdoyosarcoma is somewhat significant. Also Gbx and Nkx3.1 for prostate Ca is significant. But there are many others!
