I have a real time question for anyone out there who thinks they can help. Optimizing primer and probe concentrations are important for real-time analysis of product accumulation, ie Ct values and so on. My question is whether stringent optimization is necessary when performing only end-point analysis, in a situation where you would be interested only in identifying the presence of a product ie is it there or not?

Would optimizing primers and probes increase the sensitivity of an end-point assay?
