The replacement with valine creates a hydrophobic contact point at position 6 of the beta chain such that the deoxygenated form is prone to forming strands and aggregating.

Two main reasons for sickling or aggregates
1) As Mash says hypoxia or decrease O2 tension
2)**Increased Hemoglobin S

mjl1717.....isn't hypoxia when o2 tension is increased???
